Structural Biology and Structure-Based Drug Design in Drug Discovery

Only a very small percentage of all molecules entering drug discovery projects eventually enter phase I clinical trials. This high "attrition rate" has created extreme pressure on the pharmaceutical industry to find new and cheaper solutions in the drug discovery process. Thus, new approaches, chemical tools and technologies are being developed to save money and time in drug discovery and development.
One of these developments is related to the use of
structure-based drug design (SBDD) in drug discovery. The first examples of the successful use of this method date to the 1980ties. Since then extensive developments within the general area of structural biology, and in particular protein crystallography, proved the benefits of use of structural information and established structure-based drug design as the most superior way to follow in drug-discovery (Blundell TL et al., 2006, Philos Trans R Soc Lond B Biol Sci. 361:413-23).
